آنزیم های عسل

مقدمه:
عسل مي تواند داراي تعداد متفاوتي آنزيم باشد. بعضي از اينها بوسيله زنبورعسل تهيه مي شوند و بعضي در شهد يافت مي شوند. بر اساس انواع عسل ، منابع مختلف شهد و عسل ، آنزيمها ميزان فعاليت و واكنش متفاوتي را نشان مي دهند.
عملاً آنزيمها پروتئينهاي با ساختار پيچيده اي هستند كه موجب يك فرآيند شيم يايي ويژه مي شوند. آنها به گرما، نور مرئي و نور ماوراء بنفش و ديگر اشكال انرژي مانند ماكروويو حساس هستند.
در ميان همه آنزيمهاي عسل دياستاز و اينورتاز بيشتر مورد توجه واقع شده اند. اينها توسط زنبور به عسل افزوده مي شوند اما دليل پديدارشدن آنها در عسل تازه متفاوت است. فاكتورهايي كه حضورشان موثر است : تركيبات و اجزاء تشكيل دهنده عسل، سن زنبورها و ميزان و مقدار جريان شهد مي باشد. مثلاً جريان شديد شهد با غلظت بالا معمولاً مقادير كم دياستاز و اينورتاز را به همراه دارد

دياستاز يا آميلاز Diastase /Amilase
به بيان ساده اين آنزيم براي تبديل نشاسته به قند و دكسترين بكار مي رود و توسط زنبورها به عسل افزوده مي شود نكته اصلي جالب آن اين است كه ميزان حضورش بصورت يك نشانگر ميزان گرم شدن عسل مانند HMF است و معمولاً به همراه HMF بكار مي رود . دياستاز با يك مقياس تجربي ـ علمي بنام مقياس Gothe اندازه گيري مي شود .
بعضي عسلها بطور طبيعي داراي دياستاز پايين هستند. در استاندارد Codex حداقل 8 در مقياس Gothe منظور شده است و حد پايين براي عسلهاي دياستاز كم 3 است. در عين حال در انواع عسلهاي با دياستاز پايين ميزان HMF نبايد بيش از 10 mg/kg باشد (در مقايسه با 80 mg/kg در حالت نرمال)

اينورتاز Invertase
بر حسب مقدار فعاليتي كه انجام مي شود اينورتاز مهمترين آنزيم عسل است و اولين اهميت آن براي تبديل ساكارز به گلوكز و فروكتوز است. در حاليكه عسل رسيده ساكارز خيلي كمي دارد (معمولاً كمتر از 5%) كار اين آنزيم خيلي زود در عمر عسل انجام مي شود .

گلوكز اكسيداز Glocose Oxidase
Gox بخاطر اثري كه در خواص آنتي باكتريال عسل دارد قابل توجه است. Gox موجب تبديل گلوكز به اسيد گلوكونيك و هيدروژن پراكسايد (H2O2) مي شود كه H2o2 عامل اصلي فعاليت آنتي باكتريال بيشتر عسلها است. فعاليت Gox (معمولاً به وسيله اندازه گيري H2o2 توليد شده) در عسلهاي مختلف بسيار متفاوت است . به نظر مي رسد كه فعاليت Gox مربوط به منشاء عسل مي باشد. مثلاً در شهد ممرز (beech) معمولاً Gox سطح بالايي از فعاليت دارد.
ميزان فعاليت Gox با گرما، نور و ديگر اشكال انرژي مانند ماكروويو كاهش
مي يابد Gox به نظر مي رسد كه بيشتر توسط زنبورها به عسل افزوده مي شود. برخي دلايل موجود است كه حداقل دو نوع از اين آنزيم از قسمتهاي مختلف زنبور بدست
مي آيد و اين ممكن است تفاوت حساسيت جداگانه Gox را به نور و گرما و غيره توضيح دهد. (مثلاً صمغ اسپرگالوس يك منبع مشترك بدست آوردن Gox است ) و ممكن است بعضي از اينها در شرايط خاص توليد شوند.
Gluconic Acid اسيد اصلي يافت شده در عسل است و معمولاً براي محاسبه اسيديته عسل اندازه گيري مي شود. ايجاد گلوكونيك اسيد و H2o2 در عسل رسيده به كندي صورت مي گيرد و بيشتر توليد اين محصول در مدت زمانيكه عسل شروع به آماده شدن مي كند و توسط زنبوران عسل رطوبت از دست مي دهد انجام مي شود. اگر عسل دوباره محلول شود آنگاه اين فرآيند دوباره تسريع مي شود اين يك فاكتور مهم است كه اثرات خواص آنتي باكتريال عسل را افزايش مي دهد.
در حال حاضر ما هيچ داده منتشر شده اي درباره نيمه عمر Gox نداريم اما آنگونه كه منابع اشاره مي كنند بسيار متفاوت است
