كدام نوع از پريونها سبب بروز بيماري جنون گاوي مي*شوند

به اعتقاد محققاني كه به كشف اخير نايل آمده اند، متخصصان و پزشكان معالج از اين پس بايد با تامل بيشتري براي مداواي بيماريهايي مانند نوع انساني جنون گاويف آلزايمر و پاركينسن، اقدام كنند.

در ميان محققاني كه در مورد بيماري جنون گاوي و نوع انساني آن موسوم به "نوع تغيير يافته بيماري كروتزفلد-ياكوب (وي سي جي دي) variant "(Creutzfeldt-Jakob disease( vCJD تحقيق مي*كنند اين پرسش مطرح بوده كه كدام يك از نوع پروتئين پرايون در بروز بيماري نقش دارند.

بيماري جنون گاوي و نوع انساني آن با ازدياد شمار پروتئينهاي پرايوني كه شكلشان تغيير يافته شدت مي*يابد.

مولكولهاي بزرگتر اين نوع پرايونها بخوبي مشهود هستند اما تغيير شكل مولكولهاي كوچكتر با دشواري بيشتري مشخص مي*شود.

محققان بر اين باور بودند كه همين انواع كوچكتر از اين مولكولها هستند كه بيماري زا به شمار مي*آيند.

جي سيلوريا و همكارانش از آزمايشگاههاي راكي مانتين در هميلتن واقع در مونتانا انواع مولكولهاي تغيير شكل يافته پرايون را از مغز موشها يا همسترها آزمايشگاهي اخذ كردند و با كمك مواد پاك*كننده و ضد عفوني*كننده ، اين مولكولها را شكستند و آنها را بر اساس طولشان دسته بندي كردند.

در مرحله بعد نمونه*هاي مختلف اين سه نوع پرايون به موشهاي ديگر تزريق شد.

يك گروه از اين موشها كه زنجيره*هايي به بلندي ۲۱ پرايون به مغزشان تزريق شد پس از ۹۰ روز در اثر بيماري از پاي در آمدند. در حاليكه گروه ديگري از موشها كه زنجيره*هايي به بلندي ۳۰۰ پرايون را دريافت كردند تنها پس از آنكه ميزان غلظت محلول پرايون در مغز آنها ۷۰ برابر بيشتر از غلظت پرايون در بدن موشهاي گروه اول بود، دچار بيماري مي*شدند و پس از ۹۰ روز از پا در مي*آمدند. اين مساله روشن مي*سازد كه قابليت بيماري*زايي پرايونهاي بلند تر تا ۷۰ مرتبه كمتر است.

موشهايي كه به آنها محلولهاي با زنجيره*هاي شش تايي پرايون خورانده شد تا يكماه كاملا سالم ماندند و تنها هنگامي كه غلظت محلول تا ۶۰۰ مرتبه افزايش پيدا كرد دچار بيماري شدند.

موشهايي كه زنجيره*هايي به طول پنج پرايون يا كمتر به بدنشان عرضه شد اساسا به بيماري دچار نشدند.

روشهاي درماني كه تاكنون براي مقابله با جنون گاوي و نوع مشابه انساني آن تنظيم شده متكي به جلوگيري از رشد مولكولهاي تغيير شكل يافته پرايون در مغز است.

اما به گفته سيلوريا با تحقيقات تازه روشن مي*شود كه مي*توان از روش ديگري نيز در اين زمينه كمك گرفت. به اين معني كه اگر بتوان با احتياط زنجيره*هاي تغيير شكل يافته و متوسط يا بلند پرايون به زنجيره هاي كمتر از پنج پرايون تقليل داد، آنگاه مي*توان خاصيت بيماري زايي اين پروتئينها را از بين برد.

يافته*هاي تازه احتمالا در مورد بيماريهايي مانند آلزايمر و پاركينسن نيز كار برد دارد زيرا در اين دو بيماري نيز مولكولهاي بلند آثار بيماري زايي از خود ظاهر مي*سازند.

اما به گفته سيلوريا در شكستن اين مولكولهاي بلندتر بايد نهايت احتياط را بخرج داد تا اندازه آنها در حد مولكولهاي متوسط باقي نماند زيرا اين مولكولها خطرناك*ترين به شمار مي*آيند.
