آنتي ژن بروسلا براي تست حلقه شير

تركيب و توضيح: آنتي ژن تست حلقه شير بروسلا mrt يك سوسپانسيون از ارگانيزم كشته شده Brucella abortus رنگ آميز شده با هماتوكسيلين مي باشد كه 5/0% فنل به عنوان preservative به آن اضافه شده است اين آنتي ژن براي تشخيص وجود آنتي بادي عليه بروسلا در شير مورد استفاده قرار مي گيرد اين آنتي ژن مطابق با توصيه هاي FAO/WHO تهيه و با استانداردهاي Us Department of Agriculture Brucella Ring Test Antigen برابري مي كند.

روش آزمايشي :
1ـ يك ميلي ليتر از شير را در يك لوله آگلوتيناسيون ريخته, بطوريكه ستوني از شير به ارتفاع تقريبي 2 سانتيمتر بسازد
2- يك قطره (03/0 ميلي ليتر ) از آنتي ژن MRT به آن اضافه كنيد .
3- محتويات لوله را به مدت يك دقيقه پس ازافزودن آنتي ژن به آرامي مخلوط كنيد.
به مدت 1 الي 3 ساعت درون آنكوباتور در دماي 37 درجه سانتيگراد قرار دهيد نتايج به شرح زير ثبت مي شود:

شرائط نگهداري : در يخچال در دماي 4+ الي 8+ درجه سانتيگراد نگهداري شود از يخ زدن خودداري شود .
بسته بندي : در جعبه هاي محتوي 30 ويال 20 ميلي ليتري عرضه مي شود .

آنتي ژن بروسلا براي تست آگلوتيناسيون در لوله

تركيب و توضيح: اين سوسپانسيون از سويه 99 brocella abortus مطابق با دستورالعمل هاي كميته مشترك FAO/WHO در رابطه با بروسلا تهيه شده است. اين سوسپانسيون 10 بار غليظ شده و بايد به نسبت يك به ده در سالين 85/0% محتوي 5/0% فنل رقيق شود. وقتي كه از اين سوسپانسيون روي سرم انسان گوسفند و بز استفاده مي شود ,*ممكن است واكنشي روي دهد كه احتمالا باعث بروز اشتباه در تفسير آزمايش شود. به منظور جلوگيري از بروز اين پديده استفاده از محلول نمك 5% محتوي 5/0% فنل توصيه مي شود.

روش آزمايش: رقيق كردن سرمها به روش دوبار رقيق كردن است كه از نسبت 5: 1 شروع مي كنيم . روش كار چنين است . ابتدا 8/0 ميلي ليتر از محلول نمك محتوي 5/0% فنل را در اولين لوله آزمايش ( از سري لوله هاي آزمايش 50 يا 75 ميلي متر × 7 يا 10 ميلي متر ) ريخته و در بقيه لوله ها 5/0 ميلي ليتر مي ريزيم 2/0 ميلي ليتر از سرم را به اولين لوله افزوده محتويات آن را مخلوط كرده و 5/0 ميلي ليتر از اين مخلوط را به لوله دوم انتقال دهيد اين عمل را تا رسيدن به آخرين لوله آزمايش ادامه دهيد و پس از مخلوط كردن در لوله آخر 5/0 ميلي ليتر باقيمانده را دور بريزيد . سپس به تمام لوله ها 5/0 ميلي ليتر از سوسپانسيون آنتي ژن رقيق شده اضافه نمائيد محتويات هر لوله را با تكان دادن مخلوط كرده و لوله ها را در انكوباتور در دماي 37 درجه سانتيگراد به مدت 20 تا 24 ساعت قرار دهيد.

نتايج ثبت شده : لوله ها بايد بدون تكان خوردنم*در مقابل زمينه تيره در حاليكه منبع نور از بالا و از پشت لوله به آن مي تابد مورد آزمايش قرار گيرد.

شرايط نگهداري: در دماي 4+ الي 8+ درجه سانتيگراد ( بدون انجماد) و دور از نور خورشيد نگهداري شود.
