اشکال داروئی:
Boluses: 1250mg (as Piperazine salt)
مکانیسم اثر:
اثر نیکلوزاماید مبتنی بر وقفه در جذب گلوکز و جفت شدن (Coupling) مرحله فسفوپلاسیون اکسیداتیو در میتوکندری سلول سستودها می*باشد و به واسطه متوقف شدن سیکل کربس، اسید لاکتیک در بدن انگل تجمع یافته باعث مرگ آن می*شود.
فارماکوکینتیک:
نیکلوزاماید از مجاری گوارشی میزبان به مقدار ناچیز جذب می*شود و این خود شاید دلیل بر سمیت کم دارو باشد. این دارو به یک متابولیت غیر فعال آمینونیکلوزاماید) تبدیل می*شود که هیچ اثر شناخته شده دارویی ندارد.
موارد مصرف:
نیکلوزاماید در عفونتهای ناشی از سستودها (کرمهای نواری) در دامهای اهلی و به عنوان کشنده حلزون نیز مورد استفاده قرار می*گیرد. از این دارو برای درمان عفونتهای ناشی از تنیا پیزویفورمیس، تینا هیداتیژنا و دیپیلیدیوم کانینوم، تنیا تنیافورمیس استفاده می*شود. این دارو برای درمان عفونتهای نظیر مونیزیا در گاو، گوسفند و بز به میزان وسیعی مصرف می*شود. این دارو بر علیه انگلهای نواری بند بند مانند تنیا نوسوما در گاو، گوسفند و گوزن موثر می*باشد بطور تجربی نیکلوزاماید بر علیه عفونتهای پارانوپلو* سفالامامیلانا، آنوپلوسفالامگنا و آنوپلوسفالاپرفولیاتا در اسب و در درمان عفونتهای ناشی از سستودها نظیر بوتریسفالوس و در بعضی از انواع ماهی*ها از قبیل کول و کپور بطور موفقیت آمیزی استفاده شده است. نیکلوزاماید همچنین برای درمان عفونتهای انگلهای نواری حیوانات آزمایشگاهی (موش*ها) استفاده می*شود.
مقدار و روش مصرف:
گوسفند و بز : یک بولوس برای 15 کیلوگرم وزن بدن mg/kg) 80 ـ75)
گوساله : یک بولوس برای 20 کیلوگرم وزن بدن mg/kg) 60 ـ 50)
سگ : یک بولوس برای 10 کیلوگرم وزن بدن mg/kg) 125)
گربه : http://irvet.com/pages/drug/drug65_files/image002.gif بولوس برای 3/3 کیلو گرم وزن بدن.
زمانیکه این دارو برای درمان گوسفند و بز بکار گرفته می*شود پرهیز خاص عذایی مورد نیاز نیست ولی تجویز این دارو برای سک و گربه می*بایستی در شرایطی صورت گیرد که معده پر نباشد و 12 ساعت قبلااز تجویز دارو از دادن غذای سنگین به سگ و گربه اجتناب شود. تجویز بولوس برای گوسفند و بز با دست و یا بوسیله بولوس خوران صورت می*گیرد. تجویز برای سگ و گربه را می*توان با قرار دادن بولوس در داخل گوشت و یا خرد کردن و مخلوط کردن بولوس با مقداری غذا انجام داد.
طیور: mg/kg 200 ـ 100، ترکیب با دان، روزانه،3ـ1 روز.
حلزون*ها :WHO این دارو را به عنوان حلزون کش انتخابی برای کنترل حلزونهای حامل شیستوزوما، با غلظت 2/0 میلی گرم در هر لیتر آب به مدت 24 ساعت توصیه نموده است.
موشهای آزمایشگاهی: از راه غذا (به میزان 33/0 درصد) به مدت 7 روز پیوسته تجویز می*شود.
موارد منع مصرف:
ندارد.
تداخل داروئی:
نیکلوزاماید را می*توان همراه با چند داروی دیگر تجویز نمود. بطور مثال نیکلوزامایدو تترامیزول را می*توان به گوساله و بره برای درمان همزمان سستودها و نماتودها تجویز نمود. معمولاً از 3 گرم نیکلوزاماید و 5/0 گرم تترامیزول برای بره*ها و 6 گرم تترامیزول برای بره*ها و 6 گرم نیکلوزاماید و 1 گرم تترامیزول برای گوساله*ها استفاده می*شود.
زمان پرهیز از مصرف:
ندارد.
موارد منع مصرف:
در دامهایی که دچار انسداد مجاری قدامی دستگاه گوارش هستند نباید مصرف شود.
