اساس ژنتیکی بیماریms

در اثر بیماری مالتیپل اسکلروسیز (MS) نواحی از مغز و نخاغ دمیلینه [2] شده و این نواحی به مرور زمان گسترش می یابد . در اثر این بیماری، اغلب عصبهای بینایی ،عصبهای نخاعی و Cerebellar Conections آسیب می بینند که منجر به نوروپاتی نوری[3] ، آتاکسی و …و نهایتاً مرگ می شود. این بیماری اغلب در سنین بین 20 تا 45 سالگی بروز می کند و در بین زنان شایعتر است.
علت اصلی این بیماری هنوز مشخص نیست، اما این بیماری در اغلب موارد دارای توارث ژنتیکی می باشد و فرزندان افراد مبتلاً به MS، دارای ریسک زیادی برای ابتلا به MS می باشند . بر اساس آنالیز پیوستگی ، بین این بیماری و لوکوس هاپلوتایپ HLA که بر روی کروموزوم6 در ناحیه 6p21 قرار دارد، پیوستگی زیادی مشاهده شده است . گمان میرود این بیماری با این آنتی ژنها [4]در ارتباط باشد.بر این اساس یک ژن فرضی برای این بیماری در نظر گرفته شده است که MSSG نامیده میشود.بنظر میزسد ژن MSSG حدود 15 تا 20 سانتی مورگان با لوکوس HLA یعنی 6p21 ، فاصله داشته باشد.
بتازگی مشخص شده است که جهش در ژن PTPRC ،که بر روی کزوموزوم 1درناحیه 1q31-q32، قرار دارد ، نیز در بروز MS نقش دارد.پروتئین PTPRC یا CD45 یک زسپتور غشایی است که در غشاء همه سلولهای خونی ، بجز گلبولهای قرمز بالغ ، وجود دارد.این زسپتور ، نقش تیروزین فسفاتازی دارد ودر غیر فعال کردن JAK-کیناز ، نقش دارد.
