مقايسه اثر غلظت هاي مختلف سرم جنين گاوو سرم ميش فحل بربلوغ آزمايشگاهي تخمك هاي گوسفند

هدف از اين بررسي ارزيابي اثر غلظت هاي مختلف سرم جنين گاو و سرم ميش فحل بر بلوغ آزمايشگاهي تخمك هاي گوسفند بود. تخمدان هاي گوسفندان از دام هاي ذبح شده در كشتارگاه گرفته شده و به آزمايشگاه منتقل گرديد. در آزمايشگاه تخمك هاي موجود در داخل فوليكول هاي تخمدان با استفاده از سرنگ و روش مكش گرفته شده و پس از 4 بار شستشو در محيط كشت سلولي TC-199 حاوي مقادير معيني از دو نوع سرم به نام هاي سرم جنين گاو (Feltal calf serum) و سرم ميش فحل (Estrous sheep serum) كشت داده شد. پس از كشت به مدت 24 ساعت در گرمخانه 38.5 درجه سانتيگراد و فشار 5 درصد از گاز Co2 در هواي مرطوب، تخمك ها از سلول هاي كومولوس عاري گشته و بين لام و لامل قرار داده شده و جهت ثابت شدن در مخلوط اسيد استيك و اتانول قرار داده شدند. تخمك ها پس از مدت لازم توسط استواورسئين رنگ آميزي شده و جهت مشاهده نشانه هاي بلوغ توسط ميكروسكوپ فاز كنتراست مورد ارزيابي قرار گرفتند. اطلاعات به دست آمده توسط آزمون مربع كاي مورد ارزيابي قرار گرفته و نتايج آزمايش نشان داد كه افزودن سرم به طور معني داري (P<0.001) بلوغ تخمك ها را افزايش مي دهد. نتايج حاصل از كشت تخمك در محيط حاوي سرم ميش فحل با سرم جنين گاو قابل مقايسه مي باشد.
