شناسايي كلستريديوم سپتيكوم به روش واكنش زنجيره اي پليمراز (pcr)

هدف: تاييد تشخيص كلستريديوم سپتيكوم با استفاده از پرايمرهاي اختصاصي به روش (PCR) در بين كلستريديوم هاي جدا شده از مدفوع گوسفندان منطقه اروميه
طرح: مطالعه آزمايشگاهي
نمونه ها: 28 سويه كلستريديوم جداشده از صد نمونه مدفوع گوسفندي منطقه اروميه.
روش: نمونه برداري از مدفوع تازه گوسفندي ‘ انجام آزمايشات استاندارد ميكروب شناسي براي جداسازي كلستريديا‘ استخراج DNA از سويه ها‘ انجام PCR با استفاده از پرايمرهاي اختصاصي طراحي شده از قطعه ژني آلفا – توكسين.
نتايج: آزمون بر روي تمامي 28 سويه به اضافه سويه واكسني كلستريديوم سپتيكوم به عنوان شاهد مثبت و سه تيپ B,C,D از كلستريديوم پرفرنژنس به عنوان شاهد منفي انجام گرديد. پس ازانجام عمليات استخراج DNA‘ با پرايمر اختصاصي قطعه اي از ژن هموليزين كلستريديوم سپتيكوم (توكسين آلفاي باكتري) عمليات PCR صورت پذيرفت . هر شش سويه كلستريديوم سپتيكوم به اضافه سويه واكسن در ژل آگارزباند 270 جفت بازي را ازخود نشان دادند. كه به عنوان قطعه مورد نظر در ژن هموليزين كلستريديوم سپتيكوم تلقي گرديد.
تمامي گونه هاي ديگر كلستريدياي استفاده شده ‘ با اين پرايمر منفي بودند.
نتيجه گيري: با توجه به نتايج بدست آمده‘ مي توان پيشنهاد استفاده از روش PCR با پرايمرهاي اختصاصي براي تشخيص سريع و اختصاصي كلستريديوم سپتيكوم را توصيه نمود.
