موارد مصرف*: آمانتادين* داروي* كمكي* دردرمان* بيماري* پاركينسون* (اما نه* در علائم*خارج* هرمي* ناشي* از داروها)، بوده* و درپيشگيري* از ابتلاي* به* انواعي* از آنفلوآنزا(ضد ويروس*) سودمند است*.

مكانيسم* اثر: مكانيسم* اثر دارو مشخص*نيست*، اما ممكن* است* با تأثير بر ساخت*،آزادسازي* يا بازجذب* دوپامين*، عملكردسيستم* دوپامينرژيك* را تقويت* كند.به*عنوان* ضد ويروس* آنفلوانزا به* نظرمي*رسد از Uncoating ويروس* جلوگيري*مي*كند.

فارماكوكينتيك*: آمانتادين* از دستگاه*گوارش* جذب* مي*شود. نيمه* عمر آن* بين*2-4 ساعت* است*. بخش* عمده*داروبه*صورت* تغيير نيافته* از ادرار دفع*مي*شود.

موارد منع* مصرف*: اين* دارو در مبتلايان*به* صرع*، با سابقه* اولسرپپتيك* و درافراد با نارسايي* شديد كليه* منع* مصرف*دارد.
هشدارها:
1 ـ بر روي* اعمالي* كه* نياز به* دقت*و مهارت* دارند، مثل* رانندگي*، ممكن* است*تأثير بگذارد.
2 ـ در موارد زير با احتياط فراوان* تجويزشود:
نارسايي* كبدي* يا كليوي*، نارسايي* احتقاني*قلب*، حالات* توهم*، سالمندان*.

عوارض* جانبي*: بي*اشتهايي*، تهوع*،عصبي*شدن*، ناتواني* در تمركز، بي*خوابي*،گيجي*، تشنج*، توهمات*، گوشه*گيري*، تاري*ديد، اختلالات* گوارشي* و ادم* محيطي* ازعوارض* جانبي* دارو هستند.

نكات* قابل* توصيه*:

1 ـ از قطع* ناگهاني* دارواجتناب* شود.
2 ـ اين* دارو اثرات* ضد پاركينسوني* بسياركمي* دارد. تعداد كمي* از بيماران* از اين*
دارو سود مي*برند زيرا نسبت* به* اثرات* آن*تحمل* حاصل* مي*شود. مزيت* دارو اين*است* كه* تقريبŠ عاري* از عوارض* جانبي*جدي* است*.

مقدار مصرف*: مقدار mg/day 100 كه* بعد ازيك* هفته* به* 100 ميلي*گرم* دوبار در روزافزايش* مي*يابد، مصرف* مي*شود. دومين*دفعه* مصرف* روزانه* نبايد بعد از ساعت* 4بعد از ظهر انجام* شود. اين* دارو معمولاهمراه* ساير داروهاي* ضد پاركينسون*استفاده* مي*شود.
