بيوتكنولوژي و واكسن*هاي نسل دوم ضد انگل*هاي ليشمانيا

يك واكسن ايده*آل عليه بيماري ليشمانيوز بايد داراي چند خصوصيت باشد از جمله ايمني، كمترين تعداد ايمن*سازي توام با محافظت طولاني عليه اكثر گونه*هاي ايجادكننده ليشمانيوز، عاري بودن از محصولات حيواني، مقرون به صرفه بودن و قابليت مصرف واكسن هم براي درمان و هم براي پيشگيري.

به گزارش شماره جديد "بولتن بيوتكنولوژي"، البته ساير خصوصيات مانند تك دوز بودن و استفاده از واكسن بدون سر سوزن را نيز مي*توان در نظر گرفت.

به رغم تلاش*هاي فراواني كه در زمينه واكسن*هاي ليشمانيوز پوستي و ليشمانيوز احشايي در سطح ايران و جهان انجام گرفته است اما تاكنون واكسن ايده آلي براي آنها وارد بازار نشده است.

اما با توجه به اطلاعات و شواهد موجود كه مربوط به واكنش*هاي ميان سيستم ايمني ميزبان با انگل مي*باشد تهيه چنين واكسني غيرممكن نيست و در آينده نزديك شاهد چنين پيشرفتي خواهيم بود.

گزينه*هايي براي واكسن*هاي نسل دوم


اين كانديدها شامل ‪GP63‬ ‪ GP46‬سيستئين پروتئيناز ‪ A‬و ‪ B‬آنتي ژن*هاي ‪،LD1‬ ‪،HASPB1‬ ‪،LCR1‬ ‪،SP15‬ ‪،LEIF ،PSA-2‬ ‪ TSA ،H1‬و ‪ LCR1‬است.

واكسن ايده آل واكسني است كه حاوي چندين آنتي ژن بوده و ترجيحا از انتي ژن*هايي استفاده شود كه در ميان گونه*هاي مختلف انگل پابت باشد و در سطح آماستيگوت*ها به فراواني بيان شود.

لازم به ذكر است كه شناسايي آنتي ژن*هاي مناسب به تنهابب براي تهيه واكسن ايده آل مناسب نيست بلكه استفاده از اجوانت*هايي كه بتواند به طور قوي پاسخگوي ‪ TH1‬را تحريك كند نيز بسيار ضروري است.

اما آخرين اجونتي كه در مورد واكسن*هاي ليشمانيوز به كار گرفته شده ‪LI12‬است كه با موفقيت روبرو شده است.

جديدترين واكسن تهيه شده عليه ليشمانيورها واكنسي نوتركيب و مركب از سه آنتي ژن ‪ TSA LEIF LMST11‬است كه به نام ‪ LEISH-111F‬نامگذاري شده است و از اجوانت به كار رفته در اين واكسن ‪ IL12‬مي*باشد.

در كارآزمايي*هاي انجام گرفته بر روي حيوانات آزمايشگاهي و داوطلبان ايمني كارآيي و ايمونوژن بودن آن مورد بررسي قرار گرفته و مطلوب بوده است.

لازم به ذكر است كه آنتي ژن*هايي كه در اين واكسن به كار رفته است هم در پروماستيگوت و هم در آماستيگوت انگل وجود دارد و در ميان بيشتر گونه هاي انگل ليشمانيا از جمله ‪ L.INFANTUM‬و ‪ L.MAJOR‬كه باعث ايجاد بيماري در انسان مي*شود موجود است.

بنابراين با استفاده از اين واكسن مصونيت عليه چندگونه از انگل ايجاد مي شود و از مزاياي ديگر اين واكسن مي*توان به كاربرد همزمان آن هم براي پيشگيري و هم در درمان بيماران اشاره كرد.

نهايتا اينكه مرحله نخست كارآزمايي باليني اين واكسن در آمريكا با موفقيت به اتمام رسيده و واكسن وارد مرحله دوم كارآزمايي باليني شده و اين تنها واكسن ليشمانيوز است كه وارد مرحله دوم كارآزمايي باليني شده است.
