تهيه آنتي بيوتيك از پوست قورباغه

محققان بیوشیمی دانشگاه تربیت مدرس موفق به جداسازی و خالص سازی نوعی تركیب آنتی*بیوتیك جدید از ترشحات پوستی قورباغه*های شمال كشور شدند.

به گزارش ایسنا، تركیبات آنتی*بیوتیك استخراج شده در این طرح با توجه به مشكل مقاومت تدریجی میكروب*ها به آنتی بیوتیك*های مرسوم، كاربرد منحصر به فردی در مقابله با این قبیل میكروب*های مقاوم شده داشته و با توجه به خواص غیر همولیتیك و حفظ فعالیت ضد میكروبی آن*ها در حضور نمك*های فیزیولوژیك بدن می*توان از آن*ها در درمان موضعی عفونت*های سوختگی به ویژه عفونت*های P.aeruginosa استفاده كرد.

این تركیبات همچنین در درمان عفونت*ها و بیماری*هایی نظیر زخم پاهای بیماران دیابتی، عفونت*های دهان، عفونت*های مجاری ادراری، آكنه، ایسكمی قلبی، عفونت*های قارچی و حتی برخی سرطان*ها كاربرد خواهند داشت.

دكتر احمد آسوده، دانش*آموخته دكتری بیوشیمی دانشگاه تربیت مدرس كه با راهنمایی دكتر حسین نادری منش عضو هیات علمی دانشكده علوم پایه این دانشگاه موفق به اجرای این طرح شده است، درباره ضرورت تهیه این تركیبات آنتی بیوتیك جدید اظهار داشت: امروزه مقاومت میكروب*ها به آنتی بیوتیك*های مرسوم به یك بحران جهانی تبدیل شده به طوری كه بیش از 95 درصد سوش*های بیمارستانی s.aureuas به آنتی*بیوتیك پنی*سیلین و میتی*سیلین مقاوم شده*اند و تقریبا بیش از نیمی از عفونت*های دستگاه تنفسی در اثر میكروب*های مقاوم ایجاد می*شود؛ بنابراین شناسایی و تولید نسل جدید* آنتی بیوتیك*ها بسیار حیاتی است.

به گزارش ایسنا، وی افزود: با توجه به مصرف بالای آنتی بیوتیك*ها در ایران و بروز پدیده مقاومت میكروب*ها به آنتی*بیوتیك*ها این مساله بسیار حادتر است.

آسوده با اشاره به اهمیت ترشحات پوستی دوزیستان در تحقیقات دارویی گفت: دوزیستان به واسطه زندگی در مرداب*ها و تالاب*ها، از قویترین سیستم*های دفاع یا ایمنی اولیه برخوردارند. غدد «گرانولار» پوست دوزیستان انباری از تركیبات مختلف زیستی است. این تركیبات كه در پاسخ به محرك*های مختلف به سطح پشتی حیوان ترشح می*شود، در تنظیم نقش فیزیولوژیك پوست یا در دفاع علیه میكروارگانیسم*های مهاجم فعالیت دارند.

وی خاطرنشان كرد: برای این تحقیق بیش از صدها دوزیست Rana ridibunda یا «قورباغه مردابی» از نواحی مختلف شمال ایران (مخصوصا شهرستان گنبد) جمع آوری و ترشحات سطح پوست این دو زیست را تهیه كرده و پس از حذف پروتیین*های بالاتر از 10 كیلو دالتون، پپتیدهای كوچك كاتیونی را كه خاصیت ضد باكتریایی دارند، خالص*سازی كردیم.

به گزارش ایسنا، آسوده تصریح كرد: آنالیز پپتیدهای خالص شده نشان داد كه پپیتیدی 25 اسیدآمینه با نام بروینین 2R دارای فعالیت ضد باكتریایی علیه سوش*های استاندارد و مقاوم بیمارستانی و همچنین دارای فعالیت ضد قارچی است این پپتید تا غلظت 200 ماكروگرم بر میلی*متر، اثری بر گلبول*های قرمز نداشته و اثر آن غیر همولتیك است.

این متخصص بیوشیمی در پایان اظهار داشت: با توجه به فعالیت ضد میكروبی گسترده پپتید «بروینین 2-R» علیه سوش*های میكروبی استاندارد و مقاوم بیمارستانی و همچنین داشتن اثر ضد سرطانی و نداشتن فعالیت همولیزی می*توان با شناسایی این گونه پپتیدها نسل جدیدی از آنتی بوتیك*ها را برای جامعه پزشكی و درمانی كشور به ارمغان آورد.
